    A STUDY OF THE PKMYT1 INHIBITOR RP-6306 IN COMBINATION WITH CARBOPLATIN AND PACLITAXEL FOR TREATMENT OF RECURRENT TP53 OVARIAN AND UTERINE CANCER (GYNEREP)

    PHASE 1/2 (21-DAY CYCLE)

    6 CYCLES ONLY

    CYCLE 6

    Blood counts: All of my blood counts, including neutrophils, were very good because I did not receive Carboplatin in Cycle 5.

    CA125 decreased by 650! That's an overall decrease of 95% after 5 cycles!

    Note: I have not received a full dose of Carboplatin for the previous three infusions - 2/3 dose (Cycle 3), ½ dose (Cycle 4) and 0 dose (Cycle 5).

    Pre-meds: My pre-med schedule was modified so that I received more pre-meds and took them starting 2 days prior to infusion. 

    Infusion: There were no issues with infusion. I did not have any reaction to Carbo this round…the pre-med dosing must have done the trick!

    NEXT STEPS

    CT scan has been scheduled following this treatment.

    Study sounds good. Did you have to be platinsensitive to be included?
    I cross my finger for your CT.

    Hi! Yes, I am being treated at PMH. This was my second trial there since March '24. 

    No, I am platinum-resistant and have an over-expression of TP53. In the trial I was in, the oral trial drug RP-6306 (in combination with Carbo + Taxol) is being tested for its ability to help overcome platinum resistance. There are a few other trials underway testing this drug with other drugs…I have heard some positives about one of those trials as well. There are a couple of trials still recruiting for this drug…I haven't taken a close look at them yet but will do so today to prepare for my appointment next week. It will be interesting to see the trial reports. It certainly seems promising. The States has actually fast tracked Lunresertib (RP-6306) with Camonsertib (RP-3500) for PROC. 

    Unfortunately, the trial has ended but it brought my CA125 back down to a point I haven't been at since 2022!! I am curious to see what it is next week since I was able to have full dose Carbo at my last treatment.

    I have not yet done a comparison of my tumours from first scan in trial to the last one to see how much the tumours decreased overall. I know it was more than 30% which translates to "partial response" as per the parameters of this trial.

    I have not had such a positive response in such a short time (6 treatments over 5 months) in a very long time and it has definitely lifted my spirits.

    Thanks for your well wishes…fingers crossed 🤞. I could really use a chemo holiday right about now 🤣. The travel, accommodation and appointment scheduling has been arduous over the past year.
